Accessible LAMP-Enabled Rapid Test (ALERT) for Detecting SARS-CoV-2

The coronavirus disease 2019 (COVID-19) pandemic has highlighted bottlenecks in large-scale, frequent testing of populations for infections. Polymerase chain reaction (PCR)-based diagnostic tests are expensive, reliant on centralized labs, can take days to deliver results, and prone backlogs supply shortages. Antigen that bind detect the surface proteins a virus rapid scalable but suffer from high false negative rates. To address this problem, an inexpensive, simple, robust 60-minute do-it-yourself (DIY) workflow viral RNA nasal swabs or saliva with sensitivity (0.1 2 particles/μL) specificity (>97% true rate) utilizing reverse transcription loop-mediated isothermal amplification (RT-LAMP) was developed. ALERT (Accessible LAMP-Enabled Rapid Test) incorporates following features: (1) increased shelf-life ambient temperature storage, compared liquid mixes, by using wax layers isolate enzymes other reagents; (2) improved LAMP end-point reporting methods, sequence-specific QUASR (quenching unincorporated signal reporters); (3) sensitivity, methods without purification through use magnetic wand enable pipette-free concentration sample cell debris removal; (4) quality control nasopharyngeal-specific mRNA target; (5) co-detection respiratory viruses, such as influenza B, multiplexing QUASR-modified RT-LAMP primer sets. flexible nature allows easy, at-home point-of-care individuals higher-throughput processing labs hospitals. With minimal effort, severe acute syndrome (SARS-CoV-2)-specific sets be swapped out targets repurpose microorganisms, nucleic acid-based markers.
